A useful blister machine validation package begins before the purchase order. The user requirement specification defines the intended product, forming material, sealing system, inspection strategy, utilities, data needs and line interfaces. Those requirements become the traceability spine for design review, acceptance testing and qualification.
Qualification boundary: three activities that must not be confused
Equipment qualification evaluates facilities, systems and equipment. Process validation evaluates whether a manufacturing process can consistently deliver product meeting predetermined quality criteria. Computerised-system validation addresses software and electronic-record functions that are actually in regulated use.
A PLC or HMI does not automatically make every function subject to 21 CFR Part 11. The manufacturer must first define intended use, regulated records, electronic signatures, data flows and applicable market requirements.

1. Design Qualification: convert the URS into testable design decisions
DQ should close gaps while design changes are still inexpensive. Begin with the approved URS, then map every critical requirement to a drawing, component, software function, supplier document or planned test. A statement such as “suitable for tablets” is not testable. A useful requirement identifies the product range, blister drawing, forming material, sealing method, inspection disposition, cleaning boundary and interface conditions.
- Define products, cavity geometry, lidding and forming materials, leaflet or cartoner interfaces and expected changeovers.
- Identify critical utilities and permitted ranges at the machine connection point.
- Review product-contact materials, cleanability, access, guarding and maintenance access against the approved requirement.
- Define inspection points, reject logic, coding, batch data, user access and backup expectations before software configuration.
- Record open items, responsible owners and closure evidence in the traceability matrix.

2. Installation Qualification: verify the installed baseline
IQ establishes what was installed and which controlled documents support it. The protocol should reflect the approved design and actual site configuration. Copying a generic supplier checklist without checking utilities, drawings, instruments and interfaces leaves the manufacturer with a document set that may not represent the installed system.

3. Operational Qualification: challenge functions and ranges
OQ tests the installed equipment against approved functional requirements. For a blister line, this may include heating and forming controls, feeding, sealing, indexing, coding, inspection, reject handling, alarms, interlocks, access levels and recovery after defined interruptions. Test selection and acceptance criteria must come from process knowledge and risk assessment, not from a universal checklist.
Upper and lower operating conditions may be included where justified. “Worst case” should mean a documented, risk-based challenge inside the intended operating space. It should not be an arbitrary extreme that damages tooling or produces no useful evidence.
4. Performance Qualification: demonstrate the approved process
PQ follows successful IQ and OQ and evaluates the connected equipment under the manufacturer’s approved method and product specification. The protocol should define materials, product or justified substitute, tooling, operators, sampling, run conditions, tests, deviations and acceptance criteria. A supplier can support execution, but the manufacturer approves the protocol, assesses deviations and decides whether the evidence supports release.
Do not treat “three batches” as a universal equipment-PQ rule. Batch count and duration depend on the applicable process-validation strategy, product and process knowledge, risk, regulatory commitments and the approved protocol. FDA’s process-validation guidance describes a lifecycle approach; EU GMP Annex 15 also expects qualification and validation to be planned across the lifecycle.
Where FAT and SAT fit
Factory acceptance testing and site acceptance testing can generate useful evidence, but neither is automatically equivalent to DQ, IQ, OQ or PQ. Annex 15 allows some testing and document review at the vendor site to be used later when justified and when transport and installation do not affect the verified functionality. Your validation plan should identify what will be leveraged, repeated or supplemented on site.

The matrix should not be a decorative appendix. It should expose requirements with no design response, tests with no approved requirement, and deviations with no documented resolution.

Change control, requalification and continued control
Qualification does not end when the report is approved. Changes to tooling, forming or sealing systems, inspection logic, critical components, software configuration, utilities, products or operating ranges should enter the site’s change-control process. The impact assessment determines what document updates, testing or requalification are needed.
Routine calibration, inspection and checks should follow written programs. For US drug manufacturing, 21 CFR 211.68 addresses automatic, mechanical and electronic equipment and requires written records for calibration checks and inspections. Frequently asked questions
Who approves DQ, IQ, OQ and PQ for a blister packaging machine?
The pharmaceutical manufacturer assigns approval roles through its quality system. The supplier can prepare machine-specific evidence and support testing, but the manufacturer owns the approved requirements, protocol decisions, deviation assessment and release.
Can FAT replace OQ?
Not automatically. FAT can supply reusable evidence when the validation plan justifies it and shipping or installation does not affect the verified function. Site conditions, utilities, interfaces and functions affected by installation still require appropriate verification.
Is PQ always three consecutive batches?
No universal equipment-qualification rule fixes PQ at three batches. The approved scope should reflect the process-validation strategy, product and process knowledge, risk, regulatory commitments and applicable procedures.
Does a blister machine supplier issue a GMP validation certificate?
A supplier may provide qualification templates, test results and supporting documents. These do not replace the manufacturer’s validation system or prove that a site-specific process is validated.
When does 21 CFR Part 11 apply to a blister machine?
Applicability depends on intended use and whether the system creates, modifies, maintains, archives, retrieves or transmits regulated electronic records or uses electronic signatures. A PLC or HMI alone does not establish applicability.
